New approaches to the study of human dystrophic muscle cells in culture

摘要: Abstract Tissue culture provides a system for studying the growth and differentiation of muscle cells in controlled environment. Several studies have been carried out on diseased attempts to elucidate aetiology Duchenne muscular dystrophy (DMD) but results were equivocal. Work our laboratory recent years has yielded an improved method preparing primary cell cultures from dissociated biopsies which permits morphological biochemical evaluation these at all stages development. Our shown abnormalities behaviour, ultrastructure creatine kinase synthesis. The background is reviewed. Recently we developed cloning procedure that allows accumulation large number single selected cell. We can with this technique monitor quantitative qualitative cellular cytochemical diiferences between individual types without ambiguities inherent use mixed populations. obtained 4 different clonal preparations derived dystrophic specific features expressed by each clones respect their pattern, ultrastructure, synthesis protein surface antigen. These findings clearly illustrate potential procedures genetic expression homogeneous populations normal adult human patients X-linked disease.
