HER-2/neu gene amplification compared with HER-2/neu protein overexpression and interobserver reproducibility in invasive breast carcinoma.

摘要: We compared the detection of HER-2/ neu gene amplification by fluorescence in situ hybridization(FISH) with HER-2/neu protein overexpression immunohistochemistry using 2 antibodies on 100 archival invasive breast carcinomas.Protein for each marker was score dindependently 4 pathologists standardized criteria, and consensus results obtained from amplification. The concordance rate between FISH 76% fore2-4001 91% HercepTest. Of 37 cases positive e2-4001, 21 demonstrated no amplification; 7 24 HercepTest However, 1 61cases negative e2-4001 showed none predictive values based 0-1+, 2+, 3+immunohistochemical staining were best scored as 3+ (75% 89% HercepTest). Complete agreement among observers immunohistochemical scoring achieved 75 85 cases, respectively. pairwise kappa substantial to almost perfect Immunohistochemical maybe considered a useful screening test. While always correlated lack amplification, membranous staining, especially2+, did not predict low interobserver reproducibility separating 2+ 3+cases necessitates further confirmation before treatment decisions are made.
