The role of HLA-DR-DR and HLA-DR-DP interactions in genetic susceptibility to rheumatoid arthritis

摘要: In order ro analyze the relationships between DR and DP loci in genetic susceptibility to RA, HLA-DRB1 -DPB1 polymorphism was studied 155 RA patients compared 150 controls, using a reverse dot-blot analysis. Our data were consistent with involvement of amino acid position 71 third hypervariable region DRβ1 chain disease. The higher risk for observed who carried association lysine (K), characterizing DRB1∗ 0401 allele, an arginine (R), all other alleles (21.9% vs 0.6%, pc < 10−6, OR = 42). absence arginine, presence still associated disease (33% 19%, 0.03, 2). contrast, lysine, frequency similar controls (30% 26%, p NS). On another hand, analysis HLA-DPB1 locus showed that DPB1∗0401 allele significantly increased patient group (n 47) expressed only at β1 (82% 56% 0.008), twofold RA. results suggest role HLA-DR-DR -DR-DP interactions
