Proliferation rate but not mismatch repair affects the long-term response of colon carcinoma cells to 5FU treatment

摘要: The role of mismatch repair (MMR) in the response colon carcinoma cells to 5-fluorouracil (5FU) is not well understood. In most vitro studies only short-term was investigated. We focussed here on influence MMR status mechanism short- and long-term clinically relevant 5FU concentrations by using isogenic or semiisogenic cell line pairs expressing/nonexpressing hMLH1 protein, an important component system. show that lower survival MMR-proficient than MMR-deficient clonogenic assay due a more frequent early arrest subsequent senescence. By contrast, growth after treatment, which also affected senescence, independent from status. overall effect cumulative result proliferation rate-dependent inhibition, apoptosis necrotic death. main cytotoxic inhibition while death are minor contributions.
