摘要: Although tumor budding is linked to adverse prognosis in colorectal cancer, it remains largely unreported daily diagnostic work due the absence of a standardized scoring method. Our aim was assess inter-observer agreement novel 10-high-power-fields method for assessment at invasive front and confirm prognostic value our setting cancers. Whole tissue sections 215 cancers with full clinico-pathological follow-up information were stained cytokeratin AE1/AE3 antibody. Presence buds scored across 10-high-power fields by two pathologists additional observers asked score 50 cases randomly selected from larger cohort. The measurements correlated patient characteristics. Inter-observer correlation between observers' scores excellent (P<0.0001; intraclass coefficient=0.96). A test subgroup 65 patients (30%) used define valid cutoff high-grade remaining 70% entered into analysis. High-grade defined as an average ≥10 fields. associated higher grade (P<0.0001), TNM stage (P=0.0003), vascular invasion infiltrating border configuration (P<0.0001) reduced survival (P<0.0001). Multivariate analysis confirmed its independent effect (P=0.007) when adjusting adjuvant therapy. Using evaluating has shows agreement. Like BRE Gleason breast prostate cancers, respectively, could be basis cancer.
