Mono-(2-Ethylhexyl) Phthalate Induces Oxidative Stress and Inhibits Growth of Mouse Ovarian Antral Follicles1

摘要: Mono-(2-ethylhexyl) phthalate (MEHP) is the active metabolite of most commonly used plasticizer, di-(2-ethylhexyl) phthalate, and considered to be a reproductive toxicant. However, little known about effects MEHP on ovarian antral follicles. Thus, present study tested hypothesis that inhibits follicle growth via oxidative stress pathways. The data indicate increases reactive oxygen species (ROS) levels in follicles, whereas N-acetylcysteine (NAC; an antioxidant) restores ROS control rescues follicles from MEHP-induced inhibition growth. To further analyze mechanism by which induces growth, expression activities various key antioxidant enzymes (copper/zinc superoxide dismutase [SOD1], glutathione peroxidase [GPX], catalase [CAT]) cell-cycle regulators (Ccnd2, Ccne1, Cdk4) apoptotic (Bcl-2 Bax) were compared MEHP-treated SOD1 GPX; does not inhibit Cat expression; Ccnd2, Cdk4, Bcl-2; but Bax controls.
