Activation of p21ras by nerve growth factor in embryonic sensory neurons and PC12 cells.

摘要: p21ras is believed to be involved in the neuronal differentiation of cells responsive nerve growth factor (NGF). We show that NGF stimulates activation embryonic sensory neurons and PC12 cells. In initial 5 min exposure NGF, concentration-dependent. cells, apparent maximal was reached at 50 10 ng/ml, respectively, with half-maximal approximately 2-3 respectively. Kinetic analysis low concentrations showed slowly increases time both types while high result rapid within min. These results indicate regulates state these provides evidence suggesting signal transduction. Treatment brain-derived neurotrophic or neurotrophin-3 (NT-3) failed activate p21ras, binding alone p75LNGFR insufficient for ras activation. kinase inhibitor, K252a, which inhibits tyrosine receptor p140trk, abolished activation, p140trk major mediator by NGF.
