Adenovirally Mediated Expression of Insulin-Like Growth Factors Enhances the Function of First Trimester Placental Fibroblasts

摘要: IGFs are critical in fetal growth because of their role placental development and function. In this study, we used adenovirus (Ad-IGF) to deliver sense or antisense IGF-I IGF-II cDNA human primary fibroblasts (PPF) vitro determine whether could lead enhanced cell PPFs virally transfected with Ad-IGF-I Ad-IGF-II showed 7-fold (P < 0.01) 3-fold increases [(3)H]thymidine incorporation at 48 h post infection compared nontransfected controls. a coculture system designed assess migration, PPF cells positioned over monolayer by more than 10-fold increase migration above monolayer. After 96 culture, 2% apoptosis 16% cells, whereas 37% 25% were apoptotic. This work has established that origin amenable adenoviral transfection exert autocrine paracrine effects on proliferation, survival, suggesting enhancement IGF levels the placenta may augment function growth.
