摘要: Chitosan as a polycationic non-viral vector for gene delivery has the advantage of being biocompatible and biodegradable polymer. However, without laborious chemical modifications to its structure, it is limited use vehicle due low ability efficiently transfect under physiological conditions. To address this problem, we developed novel liposome encapsulated chitosan nanoparticles; lipochitoplexes (LCPs). nanoparticles (CsNPs) were obtained using ionic gelation technique. For purpose, an ultrapure molecular weight with high degree deacetylation was cross-linked polyanionic tripolyphosphate resulting in efficient entrapment plasmid DNA (pDNA) inside nanoparticles. LCPs prepared by incubating together anionic liposomes (DPPC/Cholesterol). The offered better pDNA protection, reduced cytotoxicity at least twofold increase transfection efficiency our also proved vivo chorioallantoic membrane model (CAM). able CAM traumatising surrounding blood vessels. This new composite system devoid modifications, organic solvents harsh production conditions makes optimal applications offering insights into field therapy.
