摘要: Abstract Ethnopharmacological relevance The medicinal mushroom Antrodia salmonea has been used as a traditional Chinese medicine and demonstrated antioxidant anti-inflammatory effects. Materials methods In the present study, we examined anti-tumor activity of fermented culture broth (AS) in vitro vivo revealed its underlying molecular mechanism action. Results Treatment human promyelocytic leukemia (HL-60) cells with AS (50–150 μg/mL) significantly reduced cell viability caused G 1 arrest via inhibition cell-cycle regulatory proteins, including cyclin D1, CDK4, E, A, phosphorylated retinoblastoma protein (p-Rb). Furthermore, treatment induced apoptosis, which was associated DNA fragmentation, followed by sequence events, intracellular ROS generation; mitochondrial dysfunction; Fas ligand activation; cytochrome c release; caspase-3, -8, -9, PARP Bcl-2/Bax dysregulation. results study suggested that AS-induced apoptosis HL-60 mediated both death receptor pathways. found effective delaying tumor incidence xenografted nude mice reducing burden. Conclusions To best our knowledge, this is first report confirming potentially beneficial against leukemia.
