Thérapie génique du glioblastome cérébral par vecteur adénoviral. Modèle expérimental chez le rat.

摘要: Resume Les effets de la therapie genique a l'aide d'adenovirus ont ete evalues dans un modele tumeur cerebrale obtenu par injection stereotaxique intracerebrale cellules gliosarcome 9L le noyau caude cerveaux rats. Sept jours apres implantation des tumorales, les adenovirus porteurs du gene β-galactosidase d'Escherichia coli (26 rats ADVbgal) ou codant pour thymidine kinase virus l'herpes (24 ADVtk) injectes facon masse tumorale selon memes reperes. 24 ayant recu l'ADVtk traites 3 l'injection virale soit intraperitoneale ganciclovir (GCV) pendant 10 jours, serum physiologique (groupe temoin). Nous avons effectue une courbe survie 15 animaux et duree moyenne etait 19 jours. Apres ADVbgal, activite trouvee tumorales chez 11 animaux. Chez suivi GCV nous observe diminution volume tumoral rapport au groupe temoin, difference moyen entre deux groupes etant significative (test Mann-Wnitney, p Ces resultats montrent l'utilite potentielle d'un HSVtk l'administration traitement glioblastomes.
