Cytochemical characterization of pituitary target cells for biotinylated gonadotropin releasing hormone.

摘要: Abstract These studies describe the application of new cytochemical stains that co-localize a biotin-labeled gonadotropin releasing hormone (GnRH) analog and FSH or LH in same field cell. Pituitary monolayer cells were stimulated with [D-Lys 6 ] GnRH labeled biotin. Biotinylated exhibited higher affinity was 7–10 × more potent than unlabeled GnRH. The avidin-biotin peroxidase complex technique (ABC) applied to localize biotinylated on use dense black substrate. Specificity tests showed stain could be eliminated by competition followed immunocytochemical for LHβ, FSHβ 25–39 ACTH different substrate (amber orange-red). Stain found surfaces 16% 60–90% stained also one gonadotropins. Most (90–100%) gonadotropes Our demonstrate binds can identified specifically as gonadotropes.
