摘要: Isolated microspore cultures from two doubled haploid (DH) lines of wheat, Triticum aestivum L., were used to develop an in vitro chromosome-doubling protocol. During the initial 24h or 48h culture microspores treated with either antimicrotubule herbicides trifluralin amiprophos-methyl (APM) concentrations ranging 0.1 μM 10 μM. Untreated control yielded 209 embryos per 100 000 microspores, which is equivalent one spike. Among regenerated plantlets 67% green, and 15% flowering plants spontaneously chromosome doubled. Treatments both had a significant effect on doubling, measured as percentage fertile regenerants. With best combination treatment duration (48 h) herbicide concentration (10 μM) among regenerants could be increased up 74% APM 65% trifluralin. The largest numbers DH spike obtained at 1-3 these resulted estimated average between genotypes 27 microspores. These results demonstrate high potential agents isolated cultures. integrated into tissue procedures will constitute efficient method for doubling future wheat breeding.
