Pressure‐Induced Activity Loss in Solid State Catalase

摘要: The pressure-induced reductions in the activities of a number enzymes solution state, and more recently solid have been reported. To further investigate effect pressure on proteins enzyme catalase was used as model. Compacts containing 150.0 +/- 0.2 mg powder were prepared instrumented laboratory presses using various compaction pressures between 0 669 MPa. After compaction, spectrophotometric assay utilized to determine pseudo-first-order rate constants for catalase-catalyzed decomposition hydrogen peroxide. These calculate change activity. Results indicated loss activity up 30% at 251 MPa or greater. While mechanism which produces is not clear, strong linear correlation seen over range (0-251 MPa) where decrease occurred. In addition, compact densities calculated correlated values. This did appear be strong.
