摘要: The requisite analyses on bone marrow biopsies are increasing: Molecular such as fluorescence in situ hybridization (FISH), polymerase chain reaction (PCR), and reverse transcriptase (RT)-PCR demanded addition to morphology immunohistochemistry improve diagnostic accuracy. Moreover, analysis of certain molecular prognostic or predictive biomarkers is increasingly mandatory the assessment hematologic diseases. In some circumstances, only formalin fixed, bone-containing tissue available for analysis. Because various fixation decalcification procedures can impair DNA RNA quality, there an urgent need standardized protocols which allow FISH PCR this study we developed a routinely applicable protocol optimize method although preserving immunohistochemical results. Therefore, compared 2 different approaches including ultrasonic versus nonultrasonic ethylenediaminetetraacetate-based reagents acid-based ones. our hands, combined use ultrasound permits successful interphase FISH, PCR, RT-PCR whereas concomitantly antigeneicity.
