摘要: This study was to examine the link between astrocyte elevated gene-1 (AEG-1) and hypoxia induced-chemoresistance in T-cell non-Hodgkin’s lymphoma (T-NHL), as well underlying molecular mechanisms. Expression of AEG-1, LC3-II, Beclin-1 were initially examined human T-NHL tissues (n = 30) normal lymph node (n = 16) using western blot, real-time PCR immunohistochemistry. Western blot also performed analyze expression cells (Hut-78 Jurkat cells) under normoxia hypoxia. Additionally, proliferation apoptosis Hut-78 exposed different concentration Adriamycin (ADM) evaluated by MTT Annexin-V FITC/PI staining assay. Finally, effects AEG-1 on ADM assessed assay, 3-MA (autophagy inhibitor) further used determine mechanism. LC3-II significantly increased compared with tissues. Incubation obviously expression. Hypoxia induced reduced ADM. overexpression decreased Moreover, inversed induced-changes cell suggested that is associated hypoxia-induced chemoresistance via regulating autophagy, uncovering a novel target against chemoresistance.
