Astrocytic AEG-1 regulates expression of TREK-1 under acute hypoxia.

摘要: TREK-1 (TWIK-related K+ channel), a member of the two-pore domain (K2P) channel family, is highly expressed in astrocytes, where it plays key role glutamate release and passive conductance. In addition, induced to protect neurons under pathological conditions such as hypoxia. However, upstream regulation remains poorly understood. this study, we found that AEG-1 (astrocyte elevated gene-1) regulates expression astrocytic hypoxic conditions. Upregulation increased knockdown dramatically decreased mRNA protein levels TREK-1, which were restored by shRNA-insensitive AEG-1. was not regulated absence AEG-1, even when HIF1α present. Together, these results suggest acts major regulator channels astrocytes SIGNIFICANCE OF THE STUDY: Previous studies have reported hypoxia increases protects neuronal cells from apoptosis. its cellular mechanism clear. study first showed mediator hypoxic-regulated normal independently HIF-1α.
