Anti-oxidative and anti-inflammatory vasoprotective effects of caloric restriction in aging: role of circulating factors and SIRT1

摘要: Endothelial dysfunction, oxidative stress and inflammation are associated with vascular aging promote the development of cardiovascular disease. Caloric restriction (CR) mitigates conditions aging, but its effects on dysfunction during remain poorly defined. To determine whether CR exerts vasoprotective in aortas ad libitum (AL) fed young aged CR-aged F344 rats were compared. Aging AL-rats was impaired acetylcholine-induced relaxation, increased NF-kappaB activity. Lifelong significantly improved endothelial function, attenuated ROS production, inhibited activity down-regulated inflammatory genes. elucidate role circulating factors mediation CR, we determined sera obtained from animals can confer anti-oxidant anti-inflammatory cultured coronary arterial cells (CAECs), mimicking CR. In CAECs presence AL serum TNFalpha elicited stress, activation gene expression. By contrast, treatment TNFalpha-induced generation prevented induction siRNA knockdown SIRT1 mitigated serum. effects, which likely mediated by factors, part, via a SIRT1-dependent pathway.
