Enzymatic oxidation of polyamines. Relationship to immunosuppressive properties.

摘要: A method for the detection of spermine oxidase activity was developed using amino acid analyzer to follow changes in composition incubation mixture or spermidine. Spermine could be detected fetal calf serum (FCS), and six major oxidation products were revealed mixture. Incubation spermidine with FCS produced which suppressed lipopolysaccharide-induced mitogenesis murine mixed lymphocyte reaction. Oxidation inhibited by inhibitors hydroxylamine isonicotinic hydrazide, inhibition immune response no longer evident suggesting that inhibitory material a product action enzyme on spermic When used as source oxidase, some suppression via cytotoxic effect not excluded. Mouse amniotic fluid found contain high levels having somewhat different specificity than FCS, this noncytotoxic inhibitor from cadaverine, spermine. These data raise questions concerning role polyamine may play immunosuppression associated pregnancy generation nonantigen-specific suppressors cells cultured other media containing activity.
