摘要: Mesenchymal cell condensation is an essential step for cartilage development. Versican/PG-M, a large chondroitin sulfate proteoglycan, one of the major molecules expressed in extracellular matrix during condensation. However, its role, especially as environment cells being condensed, has not been elucidated. Here we showed several lines evidence roles versican/PG-M chondrogenic using new chondrocytic line, N1511. Chondrogenic stimuli (treatment with parathyroid hormone, dexamethasone, 10% serum) induced marked increase transcription and protein synthesis versican/PG-M. Stable antisense clones versican/PG-M, depending on suppression expression different capacities chondrogenesis, indicated by deposition aggrecan, product. The early stages culture only V0 V1 forms, having more chains among four variants treatment those chondroitinase ABC suppressed subsequent chondrogenesis. Furthermore, β-xyloside, artificial chain initiator to consequently inhibit core proteins, In addition, forced variant V3, which no chain, disrupted organization native (V0/V1) other known be mesenchymal resulted inhibition These results suggest that involved positively regulating formation onset chondrocyte differentiation through attached chains.
