摘要: I would like to begin by explaining why turned the perfused whole organ after having used tissue slices, homogenates, mitochondria and isolated enzyme systems for over 30 years. These various preparations have proved satisfactory study of many aspects metabolism, but they are limited use in regulation metabolic processes, especially integrated control mechanisms functionally active organ. As is now known, key factors these concentrations intermediary metabolites depend on intactness structure. Metabolites diluted homogenisation, washed out slices mitochondria. This can cause major losses activities what remains may represent only a small fraction original capacity.
