摘要: 2′-Epi-2′-O-acetylthevetin B (GHSC-74), a cardiac glycoside, can be isolated from the seeds of Cerbera manghas L. We demonstrated that GHSC-74 reduced viability HepG2 cells in time- and dose-dependent manner, efficiently induced apoptosis without significantly decreasing Chang human liver Swiss albino 3T3 fibroblasts, as indicated by annexin-V/PI binding assay Hoechst 33342 staining. In addition, stimulation with series intracellular events: (1) loss mitochondrial membrane potential; (2) sustained elevation cytosolic [Ca2+]; (3) downregulation Bcl-2. BAPTA-AM, Ca2+ chelator, partly suppressed cell death prevented potential losing GHSC-74-treated cells. contrast, EGTA, an extracellular exhibited weaker effect compared to BAPTA-AM. Taken together, Ca2+-mediated pathway was found involved GHSC-74-induced apoptosis.
