摘要: Sezary syndrome (SzS), the leukemic variant of cutaneous T-cell lymphomas, is incurable. Dendritic cells (DCs) transfected with tumor mRNA can stimulate antitumor immunity in certain cancer patients. In this study, we determined whether mRNAs from could be used for loading DCs and stimulating immunity. Autologous were generated monocytes peripheral blood 10 patients SzS. Total RNA was extracted amplified by T7 vitro transcription. The induction IFN-γ granzyme B (GrB)-producing cytotoxic T lymphocytes (CTL) RNA-transfected ELISPOT assays. We found that required IL-12p70 production monocyte-derived oncogenic transcription factor Twist tyrosine kinase receptor EphA4 expressed total paired mRNAs. induced IFN-γ-producing CTLs 7 subjects GrB-producing 6 9 subjects. Both CD3+CD8+ CD4+CD25+ expanded without regulatory cells. These data support concept using a vaccine strategy requires small amounts need specific antigens
