Identification of a Novel Syntaxin- and Synaptobrevin/VAMP-binding Protein, SNAP-23, Expressed in Non-neuronal Tissues

摘要: The specificity of vesicular transport is regulated, in part, by the interaction a vesicle-associated membrane protein termed synaptobrevin/VAMP with target compartment syntaxin. These proteins, together SNAP-25 (synaptosome-associated 25 kDa), form complex which serves as binding site for general fusion machinery. Synaptobrevin/VAMP and syntaxin are ubiquitously expressed proteins believed to be involved most (if not all) cells. However, present almost exclusively brain, suggesting that homolog exists facilitate vesicle/target other tissues. Using yeast two-hybrid system, we have identified 23-kDa from human B lymphocytes (termed SNAP-23) binds tightly multiple syntaxins synaptobrevins/VAMPs vitro. SNAP-23 59% identical SNAP-25.
