PTEN-Regulated AID Transcription in Germinal Center B Cells Is Essential for the Class-Switch Recombination and IgG Antibody Responses.

摘要: Class-switch recombination (CSR) and somatic hypermutation (SHM) occur during the differentiation of germinal center B cells (GCBs). Activation-induced cytidine deaminase (AID) is responsible for both CSR SHM in GCBs. Here, we show that ablation PTEN through Cγ1-Cre mediated significantly influences responses. The GCs fail to produce IgG1 cells, high affinity antibodies nearly lost dark zone (DZ) Ptenfl/flCγ1Cre/+ mice after immunization, suggesting impaired GC structure. Further mechanistic investigations LPS- interleukin-4 stimulation induced transcription Cγ1 IgM-BCR expressing which efficiently disrupts transcription, results hyperphosphorylated AKT FoxO1 turn suppression AID transcription.
