Monoclonal antibodies against all known variants of EspA: development of a simple diagnostic test for enteropathogenic Escherichia coli based on a key virulence factor.

摘要: Enteropathogenic Escherichia coli (EPEC) are a major cause of infant diarrhoea in developing countries and significant public health issue industrialized countries. Currently there no simple tests available for the diagnosis EPEC. Serology O-antigens is widely used routinely many laboratories throughout world, even though it has been known years to be an unreliable indicator EPEC virulence. We have developed simple, low-cost immunodiagnostic test based on EspA filament, essential virulence factor related enterohaemorrhagic E. (EHEC). Using recombinant proteins five variants as immunogens, we raised panel three monoclonal antibodies mice that detects all native target bacterial cultures. The proved suitable application sandwich-type assays, including ELISA lateral flow immunoassays (LFI). Prototypes both assays were specific EHEC strains when tested against control micro-organisms. also affordable culture medium, A/E which optimizes expression allowing improved sensitivity detection compared with standard Dulbecco’s modified Eagle’s medium. Together these reagents form basis robust, informative use especially but routine screening any clinical laboratory.
