摘要: Publisher Summary This chapter reviews the Bcl-2 family, describing characteristics used for isolation of new members. The provides an overview how several homologs are isolated, cloned, and characterized. discusses functional assays to test function regulation. family proteins regulates cell death by either inducing or inhibiting apoptosis. antiapoptotic subfamily includes Bcl-2, Bcl-X L , Bcl-w, Mcl-1, A1, NR-13, CED-9. When over expressed, these protect cells from apoptotic stimuli, such as growth factor withdrawal, irradiation, DNA-damaging agents, some receptor signals. Bax Bax, Bak, Bok. BH3-only Bik, Blk, Hrk, BimL, Bad, Bid, EGL-1. Both BH3- only subfamilies, induce apoptosis when expressed. Moreover, presence conserved BH domains among members has allowed identification cloning at DNA level low stringency hybridization, PCR, database searches. By understanding differences in between future searches novel can be directed toward a specific activity.
