摘要: The present invention provides a method for mass production of human coagulation factor VII derivatives, containing: a) constructing an expression vector, containing i) nucleotide sequence dihydrofolate reductase promoter devoid at least one CCGCCC repeat from the GC-rich region thereof and encoding (DHFR) operably linked thereto, ii) cytomegalovirus (CMV) early gene derivative thereto; b) transfecting animal cell line with vector step a); c) culturing transfected in presence inhibitor to select capable expressing high efficiency; d) selected by adding group consisting sodium butyrate, vitamin K, culture medium supplement. Employing DHFR sequences, can express derivates efficiency on large scale, thus be useful preparation therapeutic agents hemophilia.
