Detection of Dicrocoelium dendriticum larval stages in mollusc and ant intermediate hosts by PCR, using mitochondrial and ribosomal internal transcribed spacer (ITS-2) sequences.

摘要: The aim of this study was to develop, perfect and validate the PCR (polymerase chain reaction) technique using mitochondrial (mt) ribosomal (ITS-2) DNA for accurate identification Dicrocoelium dendriticum in molluscs ants, first second intermediate hosts, their early detection. primers that were designed amplified a 169 pb mtDNA fragment D. permitted detection single metacercaria from Formica rufibarbis pratensis abdomen, as well brainworm head ants collected tetania. Although these did not amplify chinensis detected molluscs, they discriminate Brachylaimidae metacercariae found same mollusc. 93 bp ITS-2 is specific it chinensis, other trematodes. This very sensitive since day post-infection, only 1 abdomen ants. Both techniques are important, mainly latter.
