摘要: Fibroblast growth factor-2 (FGF-2) acts as both a potent mitogen and differentiation factor for CNS glia. In the present study, we provide evidence that intracellular cAMP determines proliferation-differentiation decision of astroglia to FGF-2 by either facilitating signalling extracellular signal-related protein kinase (ERK) or response element binding (CREB). Pharmacologically increasing levels in cultured cortical treatment with dbcAMP forskolin attenuated FGF-2-induced ERK phosphorylation glial cell proliferation. Similarly, proliferation was presence MEK inhibitor, PD98059, thus, confirming direct correlation between activation On other hand, increases prolonged CREB subsequently potentiated element-dependent transcription immediate early gene, c-fos. Moreover, effects on time-course FGF-2-dependent were mimicked suggesting cAMP-induced redirection FGF-2-signalling is linked RAF-MEK-ERK pathway.
