摘要: Abstract The following study was undertaken to determine whether an inducible calcium influx pathway is present in intact bloodstream forms of Trypanosoma brucei. Fura-2 fluorescence used demonstrate that amphiphilic peptides and amines, including melittin, mastoparan compound 48 80 , each produced a dose dependent across the plasma membrane. Calcium did not result from general disruption membrane integrity, since corresponding ethidium bromide or other divalent cations observed. Instead, selectively blocked by channel antagonists, La3+, Cd2+ Ni2+, affected Na+ tetrodotoxin amiloride. Activation trypanosome upon potential, rise intracellular free concentration ([Ca2+]i) reversed depolarization with gramicidin D. Changes Ins(1,4,5)P3 accompany influx. Overall, these data provide first evidence T. brucei, describe methods manipulate this pathway.
