摘要: We have carried out fluorescence resonance energy transfer (FRET) measurements on four-way DNA junctions in order to analyze the global structure and its dependence concentration of several types ions. A knowledge sensitivity solution environment is important for a full understanding recombination events DNA. The stereochemical arrangement four helices that make up junction was established by comparison efficiency FRET between donor acceptor molecules attached pairwise all possible permutations 5' termini duplex arms structure. conclusions are based upon series many identical which been labeled different positions, rather than determination few absolute distances. Details analysis presented, features with particular relevance structures emphasized. Three methods were employed determine FRET: (1) enhancement fluorescence, (2) decrease quantum yield, (3) shortening lifetime. results indicate arranged an antiparallel stacked X-structure when salt added solution. ion-related conformational change addition originally at low ionic strength progresses continuous noncooperative manner as increases. mode ion interaction strand exchange site discussed.
