DNA-Binding Specificity and RNA Polymerase Inhibitory Activity of Bis(aminoalkyl)anthraquinones and Bis(methylthio)vinylquinolinium Iodides

摘要: The determination of DNA-binding specificities for a series bis(methylthio)vinylquinolinium iodides and two bis(aminoalkyl)-anthraquinones was accomplished by spectral analysis, equilibrium dialysis, elevation melting temperature, inhibition DNA function as template Escherichia coli RNA-polymerase transcription activity in vitro. Studies complex formation were carried out comparison difference spectra the compounds presence native double-stranded separated-strand DNA. Base specificity interaction between demonstrated both series, particularly anthraquinones, guanine-cytosine base pair. Comparison with varied base-pair ratios showed strong preference anthraquinones pair, but quinolinium no preference. linear-binding isotherm indicated one type binding site, while types sites apparent anthraquinones. Since only anthraquinone active leukemia tests, factors other than must account antileukemic derivative.
