Gender-Divergent Profile of Bile Acid Homeostasis during Aging of Mice

摘要: Aging is a physiological process with progressive decline of adaptation and functional capacity the body. Bile acids (BAs) have been recognized as signaling molecules regulating homeostasis glucose, lipid, energy. The current study characterizes age-related changes individual BA concentrations by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) in serum liver male female C57BL/6 mice from 3 to 27 months age. Total increased 340% mice, whereas they remained relatively constant age mice. During aging, shared following changes: (1) constant; (2) proportions beta-muricholic acid (βMCA) deoxycholic (DCA) decreased between liver; (3) total BAs became more hydrophilic months. In mRNAs hepatic uptake transporters, Na(+)/taurocholate cotransporting polypeptide (Ntcp) organic anion transporting 1b2 (Oatp1b2), after 12 months, similar trends were observed for their proteins; mRNA rate-limiting enzyme synthesis, cholesterol 7α-hydroxylase (Cyp7a1), 9 high thereafter. However, Ntcp, Oatp1b2, Cyp7a1 summary, shows gender-divergent profiles composition during which likely due expression transporters Ntcp Oatp1b2 well synthetic Cyp7a1.
