摘要: Chlorozotocin, 2-[3-(2-chloroethyl)-3-nitrosoureido]-d-glucopyranose, is a newly synthesized, water-soluble nitrosourea antitumor agent that active against L1210 leukemia in mice. A 701% and 401% increase life-span were attained with dose was lethal to 10% of the animals (15 20 mg/kg, i.p.) mice treated on Day 2 or 6 tumor growth, respectively. Sixty % 2-treated 30% 6-treated survived for 90 days. At maximally effective L1210, chlorozotocin produced no significant depression normal bone marrow DNA synthesis nor peripheral neutrophil count, contrast sustained >90% inhibition ascites cell synthesis. If activity reduced toxicity are confirmed man, use this compound would facilitate treatment patients neoplastic disease who have preexisting abnormal function allow more combination anticancer agents possessing potent myelosuppressive properties.
