Evidence supporting a signal transduction pathway leading to the radiation-resistant phenotype in human tumor cells.

摘要: A signal transduction pathway, involving oncogenes and their normal counterparts the proto-oncogenes, analogous to that for cell growth differentiation has been proposed lead phenotype of cellular radioresistance (RR). In this report we provide evidence demonstrating existence such a pathway by using antisense oligonucleotides (ASO) reverse RR phenotype. Utilizing ASO directed against raf-1 gene, central component were able human tumor lines having elevated HER-2 expression or mutant form Ha-ras, two genes upstream in transduction. Additionally, anti-ras radiosensitize overexpressing cells. These results, which verify presence signaling leading RR, also have possible clinical implications use as means sensitize radioresistant tumors radiation therapy.
