摘要: MicroRNAs (miRNA) are a class of small noncoding RNAs with important posttranscriptional regulatory functions. Recent data suggest that miRNAs aberrantly expressed in many human cancers and they may play significant roles carcinogenesis. Here, we used microarrays to profile the expression 315 10 normal mucosa samples 49 stage II colon differing regard microsatellite status recurrence disease. Several were differentially between tissue tumor subtypes, miR-145 showing lowest cancer relative tissue. Microsatellite for majority could be correctly predicted based on miRNA profiles. Furthermore, biomarker profiles predict disease an overall performance accuracy 81%, indicating potential role determining aggressiveness. The levels miR-320 miR-498, both included predictive biomarker, correlated probability recurrence-free survival by multivariate analysis. We successfully verified selected using real-time reverse transcription-PCR assays mature miRNAs, whereas situ hybridization was detect accumulation epithelial cells adenocarcinoma cells. Functional studies showed potently suppressed growth three different carcinoma cell lines. In conclusion, our results perturbed numerous have functional effect behavior, and, furthermore, some prognostic clinical importance. [Cancer Res 2008;68(15):6416–24]
