Metabolite profiling and quantification of phytochemicals of Tianma–Gouteng granule in human and rat urine using ultra high performance liquid chromatography coupled with tandem mass spectrometry

摘要: Tianma-Gouteng granule has been used for the treatment of hypertension, headache, and stroke in China. However, metabolism not clear. In present study, an ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry method developed rapid identification 35 prototypes 43 metabolites human rat urine after single oral administration granule. The results showed that glucuronidation sulfation were main metabolic pathways flavonoids, alkaloids, iridoidic glycosides, anthraquinones, phenols, stilbenes found Moreover, a validated tandem was applied quantification 14 compounds (2.5 g/kg). During 0-48 h dosing, cumulative excretion rates nine prototype components 53% gastrodin, 0.07∼1.6% geniposide, baicalin baicalein, wogonoside, rhynchophylline isorhynchophylline, leonurine, emodin, indicating urinary is major way gastrodin to eliminate from body. This study provides comprehensive understanding excretive kinetics and/or rat, helpful information screening its active vivo clinical application.
