摘要: In Fluorescence Correlation Spectroscopy (FCS), fluctuations of the fluorescence intensity fluorophore-labeled molecules, excited by a sharply focused laser beam, are observed. Owing to high sensitivity, detection at level single molecules is possible. Thermodynamic can thus be revealed, which would difficult detect macroscopic level. The technique can, in principle, offer information about any molecular dynamic process nanosecond time range and longer, manifesting itself as change without need for perturbation or synchronization system studied. specific features FCS make it versatile tool biomolecular studies, including determination translational rotational diffusion, concentration density chemical kinetics, binding reactions. this review, an overview theory typical experimental setup measurements will given, along with some examples how used studies.
