A New Experimental Rat Model of Osteosarcoma Established by Intrafemoral Tumor Cell Inoculation, Useful for Biology and Therapy Investigations

摘要: Satisfactory experimental models for preclinical cancer studies must follow several criteria: (1) reproducibility of the method used to induce tumor and (2) clinical, pathological kinetic similarity with corresponding human tumors. We developed a model osteosarcoma locally induced by intrafemoral injection (OSR) cells in Sprague-Dawley rats. This yields nearly 80% bone tumors at site. These double their volume fairly slowly (in approximately 20 days) lung metastases occur 96% animals. The OSR cell-induced is characterized direct production mineralized matrix themselves, as revealed histochemical analysis. microarchitectural parameters which were quantified microscanner show an increased trabecular (+238%) when injected femur, compared controls vehicle. Osteoblastic markers such alkaline phosphatase, osteopontin, osteocalcin sialoprotein expressed vivo, whereas initially did not express some these markers, suggesting that reacquired osteoblastic phenotype favorable environment. radiological histological data this shares high similarities osteocondensing forms humans.
