Liver antioxidant and aerobic status improves after metformin and melatonin administration in a rat model of high-fat diet and mammary carcinogenesis

摘要: Oxidative stress is involved in the development of various cancers. In present study, effect long-term administration peroral antidiabetic metformin and pineal hormone melatonin on liver antioxidant aerobic status female Sprague-Dawley rats carrying mammary tumors induced by N-methyl-N-nitrosourea was evaluated. Both substances were administered a preventive curative manner (12 days before 16 weeks after carcinogen application). Carcinogen oxidative stress: level thiobarbituric acid reactive (TBARS) considered as marker oxygen species (ROS) generation increased well oxidatively modified protein content (OMP; aldehyde ketone derivates). Metformin restored succinate dehydrogenase lactate activity associated ROS production OMP to intact rats, with predominant activation superoxide dismutase (SOD) glutathione reductase (GR). Melatonin alone combination also decreased TBARS content. all groups receiving chemoprevention. The rise total capacity particularly might result from initiation anaerobic metabolism increasing SOD, GR, peroxidase activity. Long-term exerts properties liver, especially combination.
