Induction of Apoptosis in Mice with B16 Melanoma by Treatment with the Antimicrotubule Agent TZT-1027

摘要: TZT-1027, an inhibitor of tubulin polymerization, is a derivative dolastatin 10 isolated from the Indian Ocean sea hare Dolabella auricularia. TZT-1027 has good anticancer activity. We investigated effect on B16 melanoma in mice histopathologically and by agarose gel electrophoresis. At 1 mg/kg there was correlation between number mitosis-like cells apoptotic bodies. In cells, nuclear membrane had disappeared, chromosomes morphological features typical mitosis. However, not all mitosis proliferated. contrast, at 2 many bodies were detected soon after treatment, but these involved dynamic changes to cells. Furthermore, TUNEL-positive clearly visible hr treatment nuclei showed chromatin margination, cap-shape clamping, condensation electron microscopy. Therefore, we speculated that differed with dose, dose induces apoptosis tumor direct mechanism.
