摘要: BACKGROUND: White blood cell (WBC)-associated antibodies can lead to severe pulmonary transfusion reactions (transfusion-related acute lung injury [TRALI]). Investigation of a large number donor samples using the standard granulocyte immunofluorescence test (GIFT) and agglutination (GAT) proved be difficult perform due time-consuming process quantity cells required. This study describes novel flow cytometric GIFT (Flow-GIFT) method for rapid detection by analysis. STUDY DESIGN AND METHODS: A total 141 sera were analyzed presence that previously associated with suspected TRALI. As whole from human neutrophil antigen (HNA)-typed donors isolated sedimentation in ficoll density gradient. WBCs incubated respective serum binding was detected fluorescein isothiocyanate―conjugated anti-human antibody. Standard GAT performed as reference methods. RESULTS: Seven containing anti-HNA-3a, CD16, HLA Class I negative eight anti-HNA-2a, anti-CD16, anti-HLA not GAT. The Flow-GIFT able detect all antibodies, which only detectable combination In serial dilution tests, at higher dilutions than methods CONCLUSION: permits requiring fewer cells. is ideal automation will potentially open way screening population.
