Nuclear expression of the RNA-binding protein RBM3 is associated with an improved clinical outcome in breast cancer

摘要: Single-strand RNA-binding proteins (RBPs) are involved in many aspects of RNA metabolism and the regulation gene transcription. The RBP RBM3 was recently suggested to be a proto-oncogene colorectal cancer; however, such role has not been corroborated by previous studies colon or other tumor types, prognostic implications tumor-specific expression remain unclear. Mono-specific antibodies against were generated. Antibody specificity confirmed using siRNA silencing, western blotting immunohistochemistry on panel breast cancer cell lines. Using tissue microarrays IHC, protein examined 48 normal tissues 20 common cancers. Additional analysis two independent cohorts (n=1016) with long-term follow-up also carried out. upregulated compared tissues. nuclear associated low grade (P<0.001), small tumors estrogen receptor (ER) positivity (P<0.001) Ki-67 negativity both cohorts. An increased prolonged overall recurrence-free survival. value particularly pronounced hormone receptor-positive remained significant multivariate interaction after controlling for tamoxifen treatment (HR: 0.49, 95% CI: 0.30-0.79, P=0.004). These data strongly indicate that is an favorable factor cancer, seems have specific ER-positive tumors.
