摘要: We have demonstrated that the cyclohexanone method for extraction of hematin can be used to measure hemoglobin synthesis induced by erythropoietin (epo) in mouse bone marrow cells cultured medium containing methyl cellulose. The time course showed two effects due epo: an increase at day 2, which corresponded formation small erythroid colonies resulting from CFU-E (colony-forming unit, erythroid), and a very large synthesis, was maximal days 7–8 (bursts) BFU-E (burst-forming erythroid). epo dose-response curves colony counts day-2 were similar, cell-number- response these paramaters parallel. dose- curve reached plateau concentration between 3 5 units/ml, whereas 6–8-day did not reach even dose 10 units/ml.
