Characterization of an erythroid precursor cell of high proliferative capacity in normal human peripheral blood.

摘要: We have found that the peripheral blood of normal humans contains a significant number committed erythroid stem cells high proliferative capacity. These closely resemble murine burst-forming unit (BFU-E) with respect to capacity, colony morphology, and erythropoietin requirement. BFU-E were isolated from individuals by Ficoll-Hypaque density gradient centrifugation cultured in vitro using plasma culture technique. Macroscopic colonies between 100 1000 observed after 10-14 days presence either sheep or human at 0.5-4 units/ml. Individual contained 3 20 subcolonies reached maximum mean size approximately 500 cells. Colony was linearly related cell input, suggesting single cellular entity precursor each colony. The frequency capable giving rise an least per ml tested. ability assay numbers capacity should facilitate study both erythropoiesis disease states affecting which marrow samples are not available on routine basis.
