Protein folding at the air–water interface studied with x-ray reflectivity

摘要: We report the results of x-ray reflectivity measurements thin films formed by different water-soluble proteins at air–aqueous solution interface. It is demonstrated that glucose oxidase, alcohol dehydrogenase, and urease molecules denaturate interface to form 8- 14-Å-thick peptide sheets. X-ray data indicate spreading a lipid monolayer aqueous surface before protein injection does not prevent from unfolding. On other hand, crosslinking in intact enzyme layers subphase surface. A model involves interaction oxidase with phospholipid proposed. In this model, an observed decrease electron density presence explained terms “holes” film caused molecule adsorption.
