LAMA3 DNA methylation and transcriptome changes associated with chemotherapy resistance in ovarian cancer.

摘要: OBJECTIVE LAMA3 is a widely studied methylated gene in multiple tumors, but the relationship between chemotherapy resistance ovarian cancer unclear. In this study, methylation was predicted by bioinformatics, and ability of to predict prognosis confirmed experiments. METHODS Multiple databases have performed bioinformatics analysis transcription factor binding site (TFBS) on promoter region gene. Pyrosequencing detected LAMA3. QRT-PCR immunohistochemistry expression Real Time Cell Analyzer (RTCA) detects changes cell proliferation, migration invasion ability. Flow cytometry used detect apoptosis. RESULTS CPG islands 176 bp, 134 bp, 125 bp 531 bp were The 4 prediction results are basically overlapped. 7 sites predicted, one with highest score CpG island located proximal region. hypermethylation low both associated poor cancer. negatively correlated expression. After upregulation LAMA3, proliferation chemoresistant decreased, while apoptosis, increased. CONCLUSION prognosis. As typical gene, LAMA3(cg20937934) LAMA3(cg13270625) promotes invasion, apoptosis SKOV3DDP. future, mechanism will need be further studied.
