摘要: The ErbB receptor tyrosine kinases evolved as key regulatory entities enabling the extracellular milieu to communicate with intracellular machinery bring forth appropriate biological response in an ever-changing environment. Since its discovery, many aspects of family have been deciphered, emphasis on aberration signaling human diseases. However, only now, availability atomic coordinates these receptors, can we construct a comprehensive model mechanisms underlying ligand-induced dimerization and subsequent kinase activation. Furthermore, recent introduction new high-throughput screening methodologies, combined materialization systems biology perspective, reveals overwhelming network complexity, robust evolvability. This knowledge is likely impact our view diseases system perturbations resistance ErbB-targeted therapeutics manifestations robustness.
