摘要: The dynamics and environment of water in suspensions isolated rat liver mitochondria have been investigated by 1H NMR. NMR longitudinal transversal relaxation times (T1 T2) were measured the resuspension medium (2.65 s 44.57 ms) mitochondrial (1.74 23.14 s), respectively. Results showed monoexponential both cases, suggesting a fast exchange across inner membrane. Ferromagnetically induced shift extramitochondrial with nonpermeant ferromagnetic particles revealed no detectable signal from intramitochondrial compartment, confirming case. Simulations on two-compartment model indicated that residence time has an upper limit approximately 100 microseconds. Calculated apparent viscosity 30 larger than 15 cytosol erythrocytes. higher matrix could account for reductions more one order magnitude diffusion coefficient other substrates, limitations rate enzymatic reactions which are controlled favorable formation multienzyme complexes.
